We're a 30 team league that uses the Diamond Mind Baseball simulator (http://www.diamondmind.com) and projection statistics for the current season. The projections are based off of the last three seasons of stats for each player (with the most recent seasons being weighted the most heavily). There's no requirement to get the program, although it is very beneficial.
We use MLB injuries, so when a player gets injured in real life, he gets injured in the league.
We sim daily (or very close to it) following MLB's schedule (mostly, there are occasional variations).
We have 30 teams with 50 players per team. The 50 are dividied up so that each team keeps a 25 player active roster, a 15 player reserve roster of injured players, backups and prospects and a 10 player roster dedicated solely to players from the last two league rookie drafts.
We do our annual rookie draft each January (since it's slow then) and it includes any players that came into baseball during the past calendar year. That includes First time rookie draft players, Cuban defectors, Japanese players and the teenage Dominican signees.
We do place some restrictions on new GM's. We watch their trades more closely (all trades by all GM's are subject to review and approval) and there will be a vote after 6 months of membership by the full league to certify your membership as an Established GM rather than just a Newbie.
All official league decisions are made by an Executive Council of 6 GM's (4 permanent members, 2 voted in by the league as a whole). Established GM's can be voted out by the Executive Council for failing to keep up with their injured players, failing to submit a somewhat regular roster (as long as you keep up with your injured players, you'll be fine) or excessively bad behavior (being a pain in the butt isn't going to get someone tossed out, being an asshole could).
League activity means a lot to us so express yourself, have an opinion, even pick some arguments, just be aware of the line between talking some trash and making things personal or insulting (if you cross the line, someone will let you know and not in the manner of just tossing you straight out of the league).
I always advise GM's, particularly ones who have never done a sim league before, to hold off on trading for a month to see what the trade market is like. The first thing some GM's will do, is try to pull off a good deal on you (we also call this "Taking the Newbie behind the dumpster"). Just be cautious, don't feel like you HAVE to start trading right away. Take some time to look at the other teams in your division, look at how they're built, then develop a plan for your team and try to stick to it. Prospects and Pitching are both very valuable commodities in this league.

Looks good to me, though if being as asshole could get you tossed, there's a couple people that are long past being kicked out
